sgRNA target to ppc gene
sgRNA(single guide RNA) can be used to do site-specific gene editing when CRISPR/Cas9 system is present. We designed sgRNA1 target to ppc gene,which catalyzes the formation of oxaloacetic acid.
Usage and Biology
The sgRNA is composed of three regions including base-paring region which contributes to binding with target gene, dCas9 handle and terminator which are responsible for interaction with dCas9 to block target gene and tanscription termination respectively.
Ppc gene encoding the phosphoenolpyruvate carboxylase, catalyzing the conversion of phosphoenolpyruvate to oxaloacetic acid, which is one of the metabolic streams of phosphoenolpyruvate. It has been reported that inhibiting the expression of this gene can inhibit the tricarboxylic acid cycle and achieve the purpose of throttling phosphoenolpyruvate, thus improving the yield of the project target acid.
We designed sgRNA1 based on the sequence of the early coding region of this gene.
